  • Abstract In this study, closed-loop microchannel with 180° bend was designed to investigated the dynamics of DNA molecules in the microchannel, and piezoelectric diaphragm micropump was used to generated shear flow. The deformation and conformation of labeled λDNA was directly observed by fluorescence microscope, and μPIV was built to measure the velocity distribution in microchannel. This study also analyzed statistically stretch length distribution and relaxation time of DNA molecules in order to clarify the effect of Weissenberg number on the stretching of DNA molecules. The shear rate and force exerted on DNA molecules was calculated and compared with conventional stretch model. It was found that higher Wi could make the stretch length distribution more averaged, and the curved microchannel increased the shear rate.
