  • Abstract If oral delivered antigen can replace invasive vaccine treatment in grouper, it will be able to cut down labor and time cost and also improve survival rate by reducing handling stress. The objectives of this experiment are (1) to encapsule model drug (bovine serum albumin, BSA) by poly-alkylcyanoacrylate to avoid entero-degradation, (2) to improve the intestinal absorption rate by nanocapsule less than 200 nm, (3) to enhance humoral immune response by oral administrated Bacillus subtilis as adjuvant. The results showed: (1) the diameter of BSA poly-alkylcyanoacrylate nanocapsule is 80.82 ± 8.365 nm which is suitable for oral administration; (2) oral administrated BSA nanocapsule can induce BSA antibody titer increasing over time after each additional booster over 30 days period (p < 0.05); (3) BSA antibody titer is positively correlated to the dose of BSA nanocapsule (p < 0.05); (4) adding 5% B. subtilis as adjuvant can also enhance the humoral response (p < 0.05). Taken together, oral administrated BSA nanocapsule can effectively induce humoral immune response in grouper and provides the possibility for future development of oral vaccine technology.
